I currently have an E4500 and an old Asus mother board with DDR and AGP.

Is there a big difference (more than $250 or so) between building an i7 and building a strong quad core or maybe even a dual core like an E8400 or something.

I'll be building from scratch minus monitor/keyboard mouse.

Depends on what you want to do with it...

if you've got a board with an e4500, you could likely just upgrade your existing system to a q6600 or something like that. Just find the model number of your board, go under support on the asus website and they have a list of all supported cpu's.
