I was looking at Lenovo laptops and the price jump between 4gb of DIMM 2 ram and 4 gb of DIMM 1 ram was over $400. What's the difference and does it justify $400 for the same ammount of ram?

do you mean 1x dimm or 2x dimms? that would be the amount of memory modules, ie 2x 2GB modules vs 1x 4GB.

    But isn't it better to have 2 x 2gb instead of 1 x 4gb?

probably - depends on chipset. one dimm is only useful to keep a slot free for upgrades, if 4 gig is enough (which i am sure it will be) then just go with the cheaper option.

Yes. You can't benefit from the dual channel configuration with only one stick of memory.
(ie; there's a speed improvement from 2 sticks vs. 1)
