The best home defense round is .22LR or .177.
Those invaders won't know what hit 'em. Literally.
What about a 270?
Between those choices I'd say .223. smaller cartridge, lighter weight, more potential shots in a similarly-sized magazine.
Performance wise, they're both pretty potent, and have proven extremely good performance in CQC military scenarios. However, both will also grossly over-penetrate walls, and even potentially their targets.

I shot a running coyote from 150yds once with a .308 with steel sights.

I'd go with 5.56 or 6.8. Nothing is wrong with .308 but you're asking for a lot of noise and a lot of recoil.
And no shit about "I didn't ask about a a shotgun." For close range home defense I'd pick a shotgun every time.
Put a big silver silencer on it and carry it around asking people to pick a side on a coin flip...you would get some looks I bet.
I would not want to shoot either indoors without hearing protection.

But dammit as a medic, I've worked my .22LR gun shot wounds than anything else. It's like the gang bangers buy a damn brick at the start of the year to last them the next 12 months.
Not just that, but every breaking and entering that occurs in this area is met with a .22LR.
Any of them met by Ole Joe with a 22 gatling at the top of the stairs?

I've got both my 870 and AR ready for home-defense... but if SHTF in my home, i'm probably grabbing the AR. Semi auto ftmfw.

I'd still go either for a shotty or a pistol in 40 or 45.
Yeah, everyone knows 5.56 will not stop once shot until it tumbles through 5 babies and explodes killing every dog in a 3 block radius.
9mm can carry through walls just as well as .223, if not better. Nothing personal, but this gunstore myth stuff is old.
I vote .223 of the 2.
12ga slugs might be better, or a pistol sized carbine, unless you don't give a crap about your missed shot shooting through 3 layers of sheetrock, into your neighbors baby.
Seriously, though. .223 is fine. .308 is probably overkill. I'd prefer a pistol carbine for indoors, but thats just my preference.

Would it be best to shoot them in the stomach rather than the chest in self defence so it goes into the ground rather than flat shooting ?

Always CoM. Train to hit it, and target it. It is the largest target and will also has the highest probability of immediate incapacitation (other than a headshot which is hard to consisently do, especially under duress). Shooting a body part, the gun out of their hand, and other Hollywood bullshit is just that.
