I have 2 routers, Buffalo WHR-HP-54 and the Linksys WRT-54GS. They both have dd-wrt on them. The buffalo is connected to the office computer and the modem there. The linksys isn't being used right now. I have 2 other PCs that are connecting to the buffalo router by wireless g, so I want to know. What should I do/what setup should I use with the extra linksys router to give the 2 other PCs (which are upstairs) better signal and speed?

you could try an aftermarket antenna on the linksys, and put it in "bridge client" mode and run the two PCs hard-wire to it as a switch.
